OSM (Oncostatin M) is a secreted cytokine involved in homeostasis and most chronic illnesses. OSM, a member of the IL-6 (gp130) family of proteins, inhibits the proliferation of a number of tumor cell lines and plays a significant role in hematopoiesis, mesenchymal stem cell differentiation, liver regeneration, heart remodeling, nociception, inflammation and metabolism. Of all IL-6-type cytokines studied so far, OSM displays the broadest signaling profile. LIF and OSM proteins activate the JAK/STAT pathway, the MAPK protein kinases ERK1 and ERK2, the stress-activated protein kinases p38 and c-Jun N-terminal kinase (JNK), the phosphatidylinositol-3-kinase (PI3K) AKT pathway and protein kinase C delta (PRKCD). In addition, OSM exhibits many other effects by regulating the production of other cytokines including IL-6, G-CSF and GM-CSF in endothelial cells. OSM may be a tumor promoter that will provide a potential target for antiangiogenic therapy.

Cynomolgus Monkey IL-6 ELISA
Interleukin-6 (IL-6) is an interleukin that acts as both a pro-inflammatory and anti-inflammatory cytokine. It is secreted by T cells and macrophages to stimulate immune response to trauma, especially burns or other tissue damage leading to inflammation. IL-6 is also produced from muscle, and is elevated in response to muscle contraction. It is significantly elevated with exercise, and precedes the appearance of other cytokines in the circulation. Osteoblasts secrete IL-6 to stimulate osteoclast formation. Smooth muscle cells in the tunica media of many blood vessels also produce IL-6 as a pro-inflammatory cytokine. The role of IL-6 as an anti-inflammatory cytokine is mediated through its inhibitory effects on TNF-alpha and IL-1, and activation of IL-1ra and IL-10.

Chicken IL-6 Do-It-Yourself ELISA
The Chicken IL-6 Do-It-Yourself ELISA (Cat# DIY1115C-003) contains capture antibody, prottein standard, and detection antibody for development of a Chicken IL-6 ELISA. The antibodies have been determined to function in an ELISA with the standard provided. Components may also be purchased separately.

Kingfisher Biotech's Rabbit and Canine Leukemia Inhibitory Factor (LIF) recombinant proteins can be used in cell culture, as an LIF ELISA Standard, and as a Western Blot Control. Leukemia inhibitory factor, or LIF, is a member of the IL-6 family. LIF affects cell growth by inhibiting differentiation. When LIF levels drop, the cells differentiate. Leukemia inhibitory factor derives its name from its ability to induce the terminal differentiation of myeloid leukemic cells, thus preventing their continued growth. Other properties attributed to LIF include: the growth promotion and cell differentiation of different types of target cells, influence on bone metabolism, cachexia, neural development, embryogenesis and inflammation. p53 regulated LIF has been shown to facilitate implantation in the mouse model and possibly in humans. Removal of LIF pushes stem cells toward differentiation, but they retain their proliferative potential or pluripotency. LIF is typically added to stem cell culture medium to reduce spontaneous differentiation.

Deglycosylation of Bovine IL-6 (RP0014B)
Kingfisher Biotech expresses recombinant proteins in yeast so they are potentially post-translationally modified. Bovine IL-6 has a calculated molecular weight of 20.6 kDa. The Kingfisher Biotech Bovine IL-6 (RP0014B) recombinant protein was treated with deglycosylation enzymes using NEB kit Protein Deglycosylation Mix (NEB Catalog No. P6039S) to determine if the protein produced in yeast is glycosylated. The results show that Bovine IL-6 shifts in molecular weight when treated with the deglycosylation enzymes; the smear of higher MW material appears to be reduced to a single band migrating with the 22/28 kDa doublet standards. The bands showing the molecular weight of the deglycosylation enzymes are also visible. This data suggests Kingfisher Biotech's Bovine IL-6 recombinant protein produced in yeast is glycosylated.

Interleukin-6 (IL-6), once known as interferon-beta 2, has long been studied for its role in pro-inflammatory responses, notably the acute phase response and B cell maturation. Kingfisher Biotech’s Mouse IL-6 (cat # RP0380M) activity was accessed and shown to be active and comparable with other commercially available Mouse IL-6 protein. This work was done by an independent lab with expertise in bioassays. Both samples of Mouse IL-6 were reconstituted with 0.1% BSA in PBS prior to assay. Kingfisher Biotech’s Mouse IL-6 protein concentration was determined by BCA. Protein determination of the competitor’s protein is unknown. Kingfisher Biotech’s mouse IL-6 was expressed in yeast so it is naturally endotoxin-free and post-translationally modified. Mouse IL-6 purchased from Competitor A was produced in E. coli.
Method
The biological activity of Kingfisher Biotech Recombinant Mouse IL-6 was verified by proliferation of 7TD1 cells, a mouse IL-6-dependant B-cell hybridoma cell line. Briefly, 7TD1 cells were incubated with various concentrations of recombinant Mouse IL-6 protein in a dose dependent manner in complete medium containing 10% calf serum and Gentamicin for 64 hours at 37°C, 5% CO2 in a 96-well tissue culture plate. Cell proliferation was measured utilizing Promega Substrate Cell Titer 96 Aqueous One Solution Reagent and measuring the optical density of the final solution at 490 nm. The ED50 of Kingfisher Biotech Recombinant Mouse IL-6 protein was determined to be 0.006-0.009 ng/ml.

Bovine IL-6 Bioactivity
The biological activity of Kingfisher Biotech's Bovine IL-6 protein has been confirmed by an independent laboratory. Bovine and Mouse Bone Marrow-Derived Macrophages (BMDM) responded to 10 ng/mL Bovine IL-6 by up-regulating the expected IL-6 responsive genes IL4R, SOCS3 (RT-PCR) and phospho-Stat3 (Western Blot).
