Bovine CD40 Ligand ELISA

Kingfisher Biotech's Bovine CD40 Ligand Do-It-Yourself ELISA (Catalog # DIY1211B-003) contains capture antibody, protein standard, and detection antibody for development of a CD40 Ligand ELISA. The antibodies have been determined to function in an ELISA with the standard provided.

CD40 Ligand (CD40LG) is a transmembrane protein that is a member of the TNF superfamily. In addition to existing in a membrane bound form, CD40LG can also be released as a soluble molecule (sCD40LG), which has been shown to have biological functions.

CD40 interacts with CD40 ligand (aka CD40LG, TNFSF5 or CD154), which is predominantly expressed on activated CD4+ T-cells. CD40LG is transiently expressed on activated T-cells and plays a crucial role in B-cell function. Activated T-cells expressing CD40LG can also interact with CD40 on endothelial cells to induce production of inflammatory cytokines. The transient expression of CD40LG allows T-cells to stimulate selected CD40-bearing cells to participate in the immune response.

Oncostatin M Proteins

OSM (Oncostatin M) is a secreted cytokine involved in homeostasis and most chronic illnesses. OSM, a member of the IL-6 (gp130) family of proteins, inhibits the proliferation of a number of tumor cell lines and plays a significant role in hematopoiesis, mesenchymal stem cell differentiation, liver regeneration, heart remodeling, nociception, inflammation and metabolism. Of all IL-6-type cytokines studied so far, OSM displays the broadest signaling profile. LIF and OSM proteins activate the JAK/STAT pathway, the MAPK protein kinases ERK1 and ERK2, the stress-activated protein kinases p38 and c-Jun N-terminal kinase (JNK), the phosphatidylinositol-3-kinase (PI3K) AKT pathway and protein kinase C delta (PRKCD). In addition, OSM exhibits many other effects by regulating the production of other cytokines including IL-6, G-CSF and GM-CSF in endothelial cells. OSM may be a tumor promoter that will provide a potential target for antiangiogenic therapy.

C-X-C motif chemokine 10 (CXCL10) also known as Interferon gamma-induced protein 10 (IP-10) or small-inducible cytokine B10 is a member of the C-X-C chemokine family. CXCL10 (IP-10) is secreted by several cell types in response to IFN-gamma. These cell types include monocytes, endothelial cells and fibroblasts. CXCL10 (IP-10) has been attributed to several roles, such as chemoattraction for monocytes/macrophages, T cells, NK cells, and dendritic cells, promotion of T cell adhesion to endothelial cells, antitumor activity, and inhibition of bone marrow colony formation and angiogenesis.

Bovine CCL3 (MIP-1 alpha) Do-It-Yourself ELISA, ≤10 Plates

Chemokine ligand 3 (CCL3), also known as Macrophage inflammatory protein-1 alpha (MIP-1α) is a small cytokine belonging to the CC chemokine family. There are at least 27 distinct members of the C-C subgroup reported for mammals. They are characterized by two adjacent cysteines. CC chemokines induce the migration of monocytes and other cell types such as NK cells and dendritic cells. CCL3 (MIP-1α) is a chemoattractant for several different leukocytes, with varying degrees of potency (monocytes = T-cells>neutrophils>eosinophils).

VEGF-A Across Species

Kingfisher Biotech's portfolio of VEGF-A recombinant proteins includes,  bovine,  canine, equine,  feline,  mouse,  ovine,  rabbit rat,  swine, and zebrafish VEGF-A.

Vascular endothelial growth factor (VEGF) proteins stimulate vasculogenesis and angiogenesis. They are part of the system that restores the oxygen supply to tissues when blood circulation is inadequate. The normal function of VEGF proteins is to create new blood vessels during embryonic development, new blood vessels after injury, muscle following exercise, and new vessels (collateral circulation) to bypass blocked vessels. The VEGF family has six members, including VEGF-A, VEGF-B, VEGF-C, VEGF-D, VEGF-E, and Placental Growth Factor (PGF). Activity of VEGF-A, as its name implies, has been studied mostly on cells of the vascular endothelium, although it does have effects on a number of other cell types (e.g., stimulation monocyte/macrophage migration, neurons, cancer cells, kidney epithelial cells). In vitro, VEGF-A has been shown to stimulate endothelial cell mitogenesis and cell migration. VEGF-A is also a vasodilator and increases microvascular permeability and was originally referred to as vascular permeability factor (VPF).

A recent bioassay of our bovinecanine, and mouse VEGF-A based on the proliferation of HUVECs demonstrated that all three proteins are highly active on the human cell line.

Bovine IL-1 beta (IL-1F2) Do-It-Yourself ELISA

The Bovine IL-1β Do-It-Yourself ELISA (Cat# DIY1111B-003) contains capture antibody, protein standard, and detection antibody for development of a Bovine IL-1β ELISA. The antibodies have been determined to function in an ELISA with the standard provided. Components may also be purchased separately. 


The IL-1 family of cytokines encompasses eleven proteins that each share a similar β-barrel structure and bind to Ig-like receptors. Several of the well characterized members of the IL-1-like cytokines play key roles in the development and regulation of inflammation. IL-1α (IL-1F1), IL-1β (IL-1F2), and IL-18 (IL-1F4) are well-known inflammatory cytokines active in the initiation of the inflammatory reaction and in driving Th1 and Th17 inflammatory responses. In contrast, IL-1 receptor antagonist (IL-1ra; IL-1F3) and IL-36 receptor antagonist (IL-36ra; IL-1F5) reduce inflammation by blocking the binding of the agonist receptor ligands. IL-33 (IL-1F11) is thought to function as an 'alarmin' released following cell necrosis to alerting the immune system to tissue damage or stress. The biological properties of IL-37 (IL-1F7) are mainly those of down-regulating inflammation.

Bovine Leukemia Inhibitory Factor (LIF) Polyclonal Antibody Testing

Using our bovine LIF recombinant protein as the antigen, polyclonal antibodies were produced in chickens and screened for recognition of both the glycosylated and deglycosylated forms of bovine, canine, rabbit and swine LIF recombinant proteins. (Previously it was demonstrated that the Kingfisher Biotech LIF recombinant proteins were glycosylated.) The Kingfisher Biotech LIF recombinant proteins were either treated with deglycosylation enzymes or left untreated and were subsequently screened using a Western Blot. The Western Blot showed that the anti-bovine LIF polyclonal antibody recognized both the glycosylated and the deglycosylated forms of bovine (lanes 2 & 3), canine (lanes 4 & 5), rabbit (lanes 6 & 7), and swine (lanes 7 & 8) LIF recombinant proteins.

Glycosylation of Bovine, Canine, Rabbit, and Swine Leukemia Inhibitory Factor

Kingfisher Biotech expresses recombinant proteins in yeast so they are potentially post-translationally modified. Bovine, canine, rabbit and swine Leukemia Inhibitory Factor (LIF) have calculated molecular weights of  19.8, 19.7, 19.7, and 19.7 kDa, respectively. The Kingfisher Biotech LIF recombinant proteins were treated with deglycosylation enzymes using NEB kit Protein Deglycosylation Mix  (NEB Catalog No. P6039S) to determine if the protein produced in yeast is glycosylated. The results show that all four LIF recombinant proteins shifts in molecular weight when treated with the deglycosylation enzymes; the smear of higher MW material appears to be reduced to a single band migrating with the 22/28 kDa doublet standards. The bands showing the molecular weight of the deglycosylation enzymes are also visible. This data suggests Kingfisher Biotech's bovine, canine, rabbit and swine LIF recombinant protein produced in yeast are glycosylated.

Bovine Dendritic Cell Growth Cocktail

Dendritic cells are antigen-presenting cells of the immune system. Their main function is to process antigen material and present it on the cell surface to the T cells of the immune system. They act as messengers between the innate and the adaptive immune systems.
Dendritic cells are present in those tissues that are in contact with the external environment (e.g., skin, inner lining of the nose, lungs, stomach and intestines). Once activated, dendritic cells migrate to the lymph nodes where they interact with T cells and B cells to initiate and shape the adaptive immune response. During development Dendritic cells grow branched projections, the dendrites . CD209, also know as DC-SIGN, is exclusively expressed on dentritic cells in tissues, as well as on in vitro induced monocyte-derived-DC (MoDC).

IFN-gamma, or type II interferon, is a cytokine that is critical for innate and adaptive immunity against viral and intracellular bacterial infections and for tumor control. Aberrant IFN-gamma expression is associated with a number of autoinflammatory and autoimmune diseases. The importance of IFN-gamma in the immune system stems in part from its ability to inhibit viral replication directly, but, most important, derives from its immunostimulatory and immunomodulatory effects. IFN-gamma is produced predominantly by natural killer (NK) and natural killer T (NKT) cells as part of the innate immune response, and by CD4 and CD8 cytotoxic T lymphocyte (CTL) effector T cells once antigen-specific immunity develops.

Deglycosylation of Bovine IL-6 (RP0014B)

Kingfisher Biotech expresses recombinant proteins in yeast so they are potentially post-translationally modified. Bovine IL-6 has a calculated molecular weight of 20.6 kDa. The Kingfisher Biotech Bovine IL-6 (RP0014B) recombinant protein was treated with deglycosylation enzymes using NEB kit Protein Deglycosylation Mix  (NEB Catalog No. P6039S) to determine if the protein produced in yeast is glycosylated. The results show that Bovine IL-6 shifts in molecular weight when treated with the deglycosylation enzymes; the smear of higher MW material appears to be reduced to a single band migrating with the 22/28 kDa doublet standards. The bands showing the molecular weight of the deglycosylation enzymes are also visible. This data suggests Kingfisher Biotech's Bovine IL-6 recombinant protein produced in yeast is glycosylated.

Bovine Leukemia Inhibitory Factor (LIF) Recombinant Protein

Kingfisher Biotech's Bovine Leukemia Inhibitory Factor (LIF) recombinant protein can be used in cell culture, as an LIF ELISA Standard, and as a Western Blot Control. Leukemia inhibitory factor, or LIF, is a member of the IL-6 family. LIF affects cell growth by inhibiting differentiation. When LIF levels drop, the cells differentiate. Leukemia inhibitory factor derives its name from its ability to induce the terminal differentiation of myeloid leukemic cells, thus preventing their continued growth. Other properties attributed to LIF include: the growth promotion and cell differentiation of different types of target cells, influence on bone metabolism, cachexia, neural development, embryogenesis and inflammation. p53 regulated LIF has been shown to facilitate implantation in the mouse model and possibly in humans. Removal of LIF pushes stem cells toward differentiation, but they retain their proliferative potential or pluripotency.
